Project Title Development of personalized diagnosis and therapeutics for dark matters using somatic mutations in patient-derived tumor xenograft models maintaining tumor microenvironment.
Relevance medical
Description ● Dark Matter (squamous cell lung cancer, head and neck cancer, target-negative lung adenocarcinoma) cancer patient-derived PDTX establishment and clinical phenotype classification according to the response to targeted therapeutics. ● Establishment of an analysis system for profiling somatic mutations and gene expression through exome/transcriptome sequencing of patients (F0) and PDTX tumors (F2), and for discovering abnormalities in the transcriptome structure ● Biomarker validation for previously discovered target therapeutics through exome (WES) and transcriptome sequencing (RNA-seq) of PDTX ● By applying the established PDTX-genomic analysis system to the actual model of the cancer-targeted therapeutic drug, it is possible to identify the reaction mechanism of the target therapeutic drug, discover resistance predictors, and at the same time establish a precision genomic analytics that verifies previously discovered genetic markers. ● Biomarker cross-validation through Paient-Mouse Co-clinical trial ● Validation and complementation of previously discovered gene signatures that can predict the efficacy and resistance development of targeted therapeutics through analysis of the secured somatic genetic mutation-clinical factor integrated database ● Cross-validation within cohorts and verification of accuracy of predictive models through additional patient cohorts ● Establishment of biomarkers verified in PDTX as clinical grade assay and validation of patient cohorts ● PDTX-validated biomarker-based clinical trial approval (IND)
